“I think this is one of the more powerful antibiotics that has been discovered to date,” James Collins, an MIT professor of bioengineering and senior author told The Guardian . “It has remarkable activity against a broad range of antibiotic-resistant pathogens.” Beyond tests in petri-dish bacterial colonies, the team also tested halicin in mice. The antibiotic cleared up infections of a strain of bacteria resistant to all known antibiotics in a day. The team plans further study in partnership with a pharmaceutical company or nonprofit, and they hope to eventually prove it safe and effective for use in humans. This last bit remains the trickiest step, given the cost of getting a new drug approved. But Collins hopes algorithms like theirs will help. “We could dramatically reduce the cost required to get through clinical trials,” he told the Financial Times . How many novel antibiotics await discovery, and how far can AI screening take us?

The initial 6,000 compounds scanned by Barzilay and Collins’s team is a drop in the bucket.

They’ve already begun digging deeper by setting the algorithm loose on 100 million molecules from an online library of 1.5 billion compounds called the ZINC15 database. This first search took three days and turned up 23 more candidates that, like halicin, differ structurally from existing antibiotics and may be safe for humans.

Two of these—which the team will study further—appear to be especially powerful.

Even more ambitiously, Barzilay hopes the approach can find or even design novel antibiotics that kill bad bacteria with alacrity while sparing the good guys. In this way, a round of antibiotics would cure whatever ails you without taking out your whole gut microbiome in the process. All this is part of a larger movement to use machine learning algorithms in the long, expensive process of drug discovery.

Other players in the area are also training AI on the vast possibility space of drug-like compounds. Last fall, one of the leaders in the area, Insilico, was challenged by a partner to see just how fast their method could do the job. The company turned out a new a proof-of-concept drug candidate in only 46 days.

The field is still developing, however, and it has yet to be seen exactly how valuable these approaches will be in practice.

This combination medication is an antibiotic used to treat a variety of bacterial infections. It is a combination of amoxicillin, a penicillin-like antibiotic, and clavulanate, a drug that increases the effectiveness of amoxicillin.

Brand names include Amoclav®, Augmentin XR® and Clavamox®.

Studies have not been done to see if amox/clav could make it harder for a woman to become pregnant. If you were prescribed amox/clav by your healthcare provider, talk with them before making any changes in how you take this medication. It is important to consider the benefits of treating infections during pregnancy. Does taking amox/clav increase the chance of miscarriage? When taken in the recommended doses, amox/clav is unlikely to increase the chance for miscarriage.

Does taking amox/clav in the first trimester increase the chance of birth defects? In every pregnancy, a woman starts out with a 3-5% chance of having a baby with a birth defect.

Most studies have not found an increased chance for birth defects when amox/clav is taken during the first trimester; however, there are very few studies available.

There are more studies looking at the use of amoxicillin alone during pregnancy. A few studies have shown an higher chance of cleft lip (opening in the lip) with or without cleft palate (opening in the roof of the mouth) when amoxicillin is taken during the first trimester.

However there are other studies that have not found a higher chance for cleft lip with or without cleft palate.

Based on the current information, the overall chance of a cleft in lip and/or palate is considered to be low if a woman is taking amox/clav in a pregnacy.

Could taking amox/clav in the second or third trimester cause other pregnancy complications? There is limited information regarding the use of amox/clav during the second trimester, but studies so far do not report an increased chance for pregnancy problems when used in this time period. For use in the third trimester, there is one large study of women who had an increased risk for preterm labor, a condition in which a woman starts the early stages of childbirth before 37 weeks of pregnancy.

The women who were treated with amox/clav were found to have a small increased chance for a serious bacterial infection known as necrotizing enterocolitis (NEC) in their newborns. This is a condition that can injure a baby’s intestines.

Further review has found some studies that confirm this chance and others that do not.

Overall, the chance that the use penicillin and amoxicillin together of amox/clav increases the risk of NEC is low.

Does taking amox/clav in pregnancy cause any long-term problems in behavior or learning for the baby? One study that has followed hundreds of children up to age 11 years did not find that prenatal exposure to amox/clav was linked to learning or behavior problems.

Amox/clav enters the breast milk, however, based on a small number of studies; it has not been shown to cause problems for a nursing baby.

A study of 67 mothers taking this product found that there were no serious health effects in the breastfeeding infants.

While some babies had a possible allergic reaction, such as rash, diarrhea, irritability and constipation, the reactions did not last long. If you think your baby has developed a side effect from medication that might be in your milk, talk to your child’s healthcare provider.

Talk to your healthcare provider about all your breastfeeding questions. If a man takes amox/clav, could it affect his fertility (ability to get partner pregnant) or increase the chance of birth defects? There are no studies looking at possible risks to a pregnancy when the father takes amox/clav. In general, exposures that fathers have are unlikely to increase risks to a pregnancy.

What is the most important information I should know about amoxicillin and clavulanate potassium?

You should not use this medicine if you have severe kidney disease, if you have had liver problems or jaundice while taking amoxicillin and clavulanate potassium, or if you are buy amoxicillin 250mg allergic to any penicillin or cephalosporin antibiotic, such as Amoxil, Ceftin, Cefzil, Moxatag, Omnicef, and others.

Clavulanate potassium helps prevent certain bacteria from becoming resistant to amoxicillin. Amoxicillin and clavulanate potassium is a combination medicine used to treat many different infections caused by bacteria, such as sinusitis, pneumonia, ear infections, bronchitis, urinary tract infections, and infections of the skin.

Amoxicillin and clavulanate potassium may also be used for purposes not listed in this medication guide. What should I discuss with my healthcare provider before taking amoxicillin and clavulanate potassium?

You should not use this medicine if you are allergic to it, or if: you have severe kidney disease (or if you are on dialysis); you have had liver problems or jaundice while taking amoxicillin and clavulanate potassium; or you are allergic to any penicillin or cephalosporin antibiotic, such as Amoxil, Ceftin, Cefzil, Moxatag, Omnicef, and others.

Tell your doctor if you have ever had: liver disease (hepatitis or jaundice); kidney disease; or mononucleosis.

The liquid or chewable tablet may contain phenylalanine. Tell your doctor if you have phenylketonuria (PKU).

Tell your doctor if you are pregnant or breastfeeding.

Amoxicillin and clavulanate potassium can make birth control pills less effective.

Ask your doctor about using a non-hormonal birth control (condom, diaphragm, cervical cap, or contraceptive sponge) to prevent pregnancy.

Do not give this medicine to a child without medical advice.

How should I take amoxicillin and clavulanate potassium?

Follow all directions on your prescription label and read all medication guides or instruction sheets. Amoxicillin and clavulanate potassium may work best if you take it at the start of a meal. Swallow the pill whole, or break the pill in half and take both halves one at a time. Tell your doctor if you have trouble swallowing a whole or half pill. You must chew the chewable tablet before you swallow it. Shake the oral suspension (liquid) before you measure a dose. Use the dosing syringe provided, or use a medicine dose-measuring device (not a kitchen spoon).

This medicine can affect the results of certain medical tests.

Tell any doctor who treats you that you are using amoxicillin and clavulanate potassium. Use this medicine for the full prescribed length of time, even if your symptoms quickly improve. Skipping doses can increase your risk of infection that is resistant to medication.

Amoxicillin and clavulanate potassium will not treat a viral infection such as the flu or a common cold.

Store the tablets at room temperature away from moisture and heat. Take the medicine as soon as you can, but skip the missed dose if it is almost time for your next dose. Seek emergency medical attention or call the Poison Help line at 1-800-222-1222.

Overdose can cause nausea, vomiting, stomach pain, diarrhea, skin rash, drowsiness, hyperactivity, and decreased urination. What should I avoid while taking amoxicillin and clavulanate potassium?

Avoid taking this medicine together with or just after eating a high-fat meal. This will make it harder for your body to absorb the medication. Antibiotic medicines can cause diarrhea, which may be a sign of a new infection. If you have diarrhea that is watery or bloody, call your doctor before using anti-diarrhea medicine. What are the possible side effects of amoxicillin and clavulanate potassium? Get emergency medical help if you have signs of an allergic reaction (hives, difficult breathing, swelling in your face or throat) or a severe skin reaction (fever, sore throat, burning eyes, skin pain, red or purple skin rash with blistering and peeling).

Call your doctor at once if you have: severe stomach pain, diarrhea that is watery or bloody (even if it occurs months after your last dose); pale or yellowed skin, dark colored urine, fever, confusion or weakness; loss of appetite, upper stomach pain; little or no urination; or easy bruising or bleeding.

Common side effects may include: nausea, vomiting; diarrhea; rash, itching; vaginal itching or discharge; or diaper rash.
